New approach to reverse multiple sclerosis in mice models

ScienceDaily (June 28, 2012) ? Mayo Clinic researchers have successfully used smaller, folded DNA molecules to stimulate regeneration and repair of nerve coatings in mice that mimic multiple sclerosis (MS). They say the finding, published June 28 in the journal PLoS ONE, suggests new possible therapies for MS patients.

"The problem has been to find a way to encourage the nervous system to regenerate its own myelin (the coating on the nerves) so nerve cells can recover from an MS attack," says L. James Maher III, Ph.D., Mayo Clinic biochemist and senior author on the paper. "We show here that these small molecules, called aptamers, can stimulate repair in the mice we are studying."

More than 200,000 people have multiple sclerosis. There is no cure and no effective therapy to stop progression or repair damage to the myelin sheath that surrounds and protects the nerves. Without that protection, nerve fibers will be damaged, leading to declining mobility and cognitive function, and other debilitating complications.

MS researchers, including Mayo neurologist Moses Rodriguez, M.D., a co-author on this paper, have focused on monoclonal antibodies in mice to stimulate myelin repair. The Rodriguez and Maher teams, working together, have determined that the aptamers are not only effective, but they are easy and cheap to synthesize -- an important point for drug developers. They also are stable and not likely to cause an immune response. This new approach must be validated in other mouse models to see if it might be a candidate for human clinical trials.

The monoclonal antibodies used in earlier research are large and complex, but were shown to promote both cell signaling and remyelination of central nervous system lesions in mice. The aptamers used in this study are less than one-tenth the size of antibodies and are single-strands of DNA containing only 40 nucleotide units.

The research was supported by Mayo Clinic and the National Multiple Sclerosis Society. Co-authors include Branislav Nastasijevic, Brent Wright, Ph.D., John Smestad, and Arthur Warrington, Ph.D., all of Mayo Clinic.

Ann Curry Confirms She Is Leaving 'Today'

Ann Curry confirmed early on Thursday morning that she is leaving the "Today" show after little more than a year as co-host.

Speaking to USA Today, Curry said that she would announce to viewers on Thursday's program that her tenure has come to an end.

She said it was "going to be a bit of a tough day" when she broke the news to the audience.

"I'm going to have to tell our viewers," she said. "That's what makes me more emotional than anything. I don't want to leave them. I love them. And I will really miss them."

The paper wrote that Curry has signed a long-term deal with NBC News. She will become a roving national and international correspondent, producing specials and covering the more serious journalism which she has long said comes easiest to her.

However, the announcement brings a swift, sad finish to Curry's tenure as a co-host of the iconic morning show. It was a job she had coveted deeply, and had waited to grab a hold of even as she was passed over once before in favor of Meredith Vieira. But falling ratings and persistent rumblings that she is not the right fit for the job have led NBC to push her out of her position.

It is not yet known exactly what kind of farewell the show will give her. Savannah Guthrie, one of the co-hosts of the third hour, has reportedly already been asked to replace Curry.

In the USA Today interview, Curry struck a somewhat defiant note. She rejected the idea that she had bad chemistry with co-host Matt Lauer, and, when asked if she thought she had been given enough time to settle into her new role after fourteen years as the show's newsreader, said, "No, I do not."

She also said that she had been ready to leave NBC entirely, having been "hurt deeply" by the protracted ugliness of the past week. Since news broke last Wednesday of her impending departure, her every move has been tracked by a cloud of press attention. One report began, "Ann Curry showed up again for work." (NBC maintained a stony silence throughout.)

But Curry said that NBC News president Steve Capus convinced her to stay, offering a job that she described as rich with opportunities for hard news reporting.

It has been widely rumored that she will be making up to $10 million a year with the network. Curry said this was not true.

The brutal conclusion to Curry's hosting career highlights just how important maintaining the supremacy of "Today" is to NBC. The show makes hundreds of millions of dollars in profits every year, but that money was imperiled by the ascendancy of ABC rival "Good Morning America" in the ratings. When that show officially snapped the 16-year "Today" ratings winning streak, media watchers immediately began wondering how NBC would respond. Unfortunately for Curry, the response has cost her what she has called her "dream job."

George Zimmerman "Missed Opportunities" to Defuse Trayvon Martin Confrontation, Investigator Finds

George Zimmerman failed to identify himself twice and missed opportunities to defuse a confrontation with Trayvon Martin, a detective says in a newly released report.

Zimmerman, a neighborhood watch volunteer, is charged with second-degree murder in the February 26 shooting death of Martin, 17, in Sanford, Florida.

The report, which Florida prosecutors released Tuesday, includes a video of Zimmerman showing injuries he said he suffered in the altercation with Martin.

Zimmerman, 28, told police he shot the teen in self-defense, but many feel he racially profiled Martin and ignored a 911 dispatcher's advice not to follow him.

Among the documents released was a report from a police officer who conducted computerized voice stress analysis tests on Zimmerman following the incident.

"Mr. Zimmerman was subjected to two exams, and was found to be classified as No Deception Indicated," the report said.

Also released was an unredacted capias request, a request that someone be taken into custody, prepared by the lead police investigator in the case.

Christopher Serino wrote in the report:

"Investigative findings show that (Zimmerman) had at least two opportunities to speak with (Martin) in order to defuse the circumstances surrounding their encounter."

"On at least two occasions (Zimmerman) failed to identify himself as a concerned resident or a neighborhood watch volunteer."

The detective also said Zimmerman's actions "are inconsistent with those of a person who has stated he was in fear of another subject."

In the same report, Serino wrote that Zimmerman's injuries were "marginally consistent with a life-threatening violent episode as described by him."

The report is one of several key pieces of information, including the Trayvon Martin autopsy report, to be released in advance of the trial.

Zimmerman's defense attorney, Mark O'Mara, filed a motion Wednesday asking that his client be allowed to appear at a Friday bond hearing in "civilian clothing instead of a prison uniform and without restraints."

Zimmerman's bail was revoked this month after after a judge said he misled the court about his finances. He had been released in April.

Video and photos of Zimmerman shackled and wearing jail garb could prejudice prospective jurors, which is a serious concern, O'Mara said.

In response to the document release, O'Mara said Tuesday it will be up to a judge and jury to interpret what Serino's report means to the case.

"His suggestion that it's marginally consistent, again, is up for review. Those people will have to look at it, whether it'd be Judge (Kenneth) Lester in a motion hearing where the jury will also make that determination," he said.

He said a jury will decide "whether or not those injuries give rise to a reasonable belief in George's mind that he was a victim of great bodily injury or death."

Martin's family attorney, Benjamin Crump, said he agrees with the detective.

"I am disappointed because I think public as whole expects integrity in what our public records share to the public. If that information is mis-labeled than that should be reflected on the public domain,? says Edwards.

Susan Simmons, chairman of the board of equalization echoes a need for change.

"They need to have the numbers there correct as other localities do,? says Simmons.

The Board of Supervisors says they know the numbers are wrong, and need to be corrected. One idea involves a software upgrade similar to what other localities use. Nothing definite until the taxation department comes back with its findings in two weeks.

"I believe if we need to correct them in large numbers we will do it. I am going to stand in there and fight to the end because what is right is right wrong is wrong, but when you see glaring information spread over the county of 96,000 people, if you don't wake up and see something is smoking in the kitchen than something is wrong,? says Church.

Supervisor Butch Church says at this point, they do not know how many people overpaid in taxes. The hope is to get that information from the taxation department.

Seamus was loaded into Dwayne's borrowed trailer.? Right now he is on his way to Smithville where he will be processed.? The emotions behind this decision are, obviously, complicated and raw.? I wrote at length about my initial decision to process one of our steers here and little of what was said then has changed for me now.

But today was much, much harder for me than butchering Rooney last year.? I've been sleepless for a week and wrestled with the knowledge that just as easily as I decided we would do this, I could decide not to.? That sort of power is frightening.? Although I was positive I wouldn't be able to help load him, I managed to calmly participate in what ended up being a very smooth loading from round pen to trailer.? While holding a bucket of feed over the fence to lure him forward, my upper arm snagged on the barbed wire.? There's a deep, open hole there that will probably scar.? It seems appropriate.

I came back inside and poured a tall glass of iced tea, sat down at the computer and wrote a note to my sister - if we all had to do this each time we wanted a hamburger, our diets would be dramatically different.? BBQ ribs would be more of a luxury and a blessing.? There's nothing sterile or simple about consuming meat.? I was horrified, too, when we first discussed the fact that Matilda needed a new home, the reality of hay prices, the fact that this animal was always intended for slaughter.? I broke down twice.? I cried when he was driven away, then I came inside and washed my gouged arm and silently thanked whatever forces came together to allow us this piece of dirt and these animals.

Many of our choices around here regarding animals are heavy.? They should be.? This is not the last animal we will raise for food - for us, our families and our friends.? This isn't the last time I'll lean against the kitchen counter allowing myself two whole minutes of quiet tears.? Like last year, I'm full with gratitude for his time here on the farm and his contribution as food for this table and others.??

Age is just a number... except when it comes to marriage.

Let's look at my stats:
Current age - 29
Divorced for - 8 months
Separated for - 1 year, 9 months
Age when I met my ex - 19
Age when I married - 24

Which brings me to my point: couples should not be allowed to get married before age 25.

While I know that this statement is going to make me very unpopular with readers, I do believe that it would be for the best -- better both for the institution of marriage and the individuals getting married -- if we could change the law to prevent couples from getting married before the age of 25.

In my experience, marriage before 25 was not the smartest idea. I met my ex at the tender young age of 19 (just a few months after my birthday). I was enjoying the freedom of drinking and partying legally for the first time (I live in Canada where the drinking age is 19). I had yet to figure out who I was or what I wanted in my life. I was na?ve and impressionable, and when I met my much older ex, I was perfectly happy to let him take control of my life, creating a relationship dynamic that continued for the nine years we were together.

We decided to get married when I was 24. It seemed like the right thing to do at the time because everyone knows that after five years, you should be married or at least engaged, right? It was definitely the message that I was getting from all of our friends. So we took the plunge, getting married on the beach in Miami Beach in front of our friends and family.

That's where the problems began. Once the excitement of planning a beach wedding was over, after the suntan had faded, I was left simply living my life with my husband. Don't get me wrong, I loved him -- and a part of me always will. But because we had started dating at such a young age, he was marrying someone who had absolutely no idea who she was and what she wanted in her life. In short, it was a recipe for divorce.

People under the age of 25 are still discovering themselves; they are figuring out what is most important in their lives. They are discovering the joys (and heartache) of being in a relationship, and then the partying that often characterizes life between relationships. They are figuring out what their relationship "deal-breakers" are and who their most appropriate partners would be. While a person may be 100 percent certain that they love something -- or someone -- at the age of 21, by 29, they will most likely completely change their mind. Life is anything but certain.

My opinions are based solely on my personal experiences and the experiences of the people that I know and have observed. That being said, marriage and divorce statistics do support my claim. According to the National Center for Health Statistics, approximately 60 percent of marriages in which the couple marries between age 20 and 25 will end in divorce. A scary figure for young lovebirds... but definitely one that validates my opinion.

Who knows? Maybe there are 20-year-olds that get married and stay madly in love for their whole lives. Maybe puppy love can last forever.

Could be. Maybe there is such thing as fairies and unicorns too.

Employers are slowing down their hiring. Consumers have turned cautious. It's not the present state of affairs that's got them worried, it's what lies ahead: the potential breakup of the eurozone, the uncertainty around US elections, and the coming expiration of US tax cuts.

"We hear that up and down the food chain," says Jim John, chief operating officer of?Beyond.com, an online career network based in?King of Prussia, Pa., for employers and job-seekers. "Employers are pulling in their horns."

The latest employment report, released Friday, confirm the jitters. Employers added only 69,000 jobs in May, the Department of Labor reported, the smallest increase in a year and less than half what a consensus of economists had forecast. Employment gains for the two previous months were also adjusted downward by a combined 49,000 jobs. The unemployment rate ticked up from 8.1 percent to 8.2 percent.

For the past two months, economists have speculated that the weaker-than-expected jobs numbers were a kind of "payback" for the unusually strong employment gains in the winter, caused by unusually warm weather that boosted everything from construction work to car sales. Now, however, they're acknowledging that other factors are at work.

"It does now appear that the global slowdown, and events in Europe particularly, are beginning to have a more marked impact on the US economy," writes Paul Ashworth, chief US economist at Toronto-based Capital Economics, in an analysis of Friday's numbers.

"The latest figures cast doubt on whether the economy has enough momentum to achieve even the 2.2 percent growth rate we had expected for this year," writes Nigel Gault, chief US economist at IHS Global Insight in Lexington, Mass., in a research note. "Given the uncertainties over the eurozone crisis, emerging market growth, the US elections and the 'fiscal cliff,' there are plenty of reasons for businesses to stay cautious in their hiring plans, even if surging gasoline prices are for the moment no longer on the list of things to worry about."

The weakness in hiring appears to be centered among large companies, says Mr. John. Since March, Fortune 100 companies in retail, financial services, and communications began advertising fewer job openings on Beyond.com. The good news is that the number of jobs posted on the company's websites are more than double the level of a year ago. But the slowdown in job ads from big companies continues to intensify, a trend he expects to last for several more months.

"The [large] employers are saying: 'Let's wait and see,' " he says, as uncertainty mounts about Europe, the outcome of the US elections, and tax policy. That could lead to a mild recession. "We might talk ourselves into negative growth for a quarter or two," he cautions.

Even before the unemployment numbers came out, former Fed Chairman Alan Greenspan was sounding a cautious note. "There is a fear of the future," he told CNBC Friday, "and when you begin to try to disaggregate?what's causing that, you come up with probably 40 percent of it is the fact that the economy is sagging."

Scientists at the University of York and GlaxoSmithKline (GSK) Australia have discovered a complex gene cluster responsible for the synthesis of the medicinal compound noscapine.

The discovery, published in the latest issue of Science, reveals that the pathway for synthesis of noscapine is controlled by a complex cluster of ten genes encoding five different enzyme classes. This is the most complex gene cluster ever found in plants. The discovery has revealed the previously unknown biochemical pathway for noscapine synthesis. It will also greatly accelerate the breeding of high-noscapine poppy varieties.

The breakthrough came when the scientists discovered that poppy varieties that produce noscapine express a number of genes that are absent in varieties that are noscapine free. They then analysed the inheritance pattern of these genes in hundreds of offspring from crosses between noscapine and no-noscapine varieties. When they saw that all of these genes are inherited together, they realised they could be looking at an incredibly complex gene cluster. The identity and arrangement of genes in the cluster was determined by cloning and DNA sequencing.

Professor Ian Graham, Director of the Centre for Novel Agricultural Products at the University of York, said: "We were amazed to find that this gene cluster encodes for almost the entire biosynthetic pathway for noscapine. With this one discovery we have been able to produce an outline of the pathway and define a number of the steps involved - something that normally takes years."

Noscapine has been used as a suppressant in cough mixtures for decades. More recently, it has been reported to have anti-cancer activity and is currently in early stage clinical trials. Noscapine was one of the first natural products to be chemically characterised almost 200 years ago. Until now, however, the pathway for production and the genes involved in its synthesis have not been identified.

Tim Bowser, Head of R&D for GSK Australia's Opiates Division, said: "The fact that the genes are grouped in a cluster means that plant breeding becomes faster and easier. GSK are using this discovery to develop high yielding commercial noscapine poppies in order to establish a reliable route of supply."

Commercial poppy farming traditionally involved lancing the seed capsule to release the latex from which medicinal alkaloids, including noscapine, could be extracted. Since the 1960s commercial industrial techniques have largely switched to large scale poppy harvesting and extraction focussed on other medicinal poppy alkaloids like morphine and thebaine used to manufacture pain medicines. This trend has developed at the expense of noscapine which until now was only available from traditional techniques.
